چکیده

The influence of in utero low-dose ionizing radiation exposure on murine hemopoietic embryogenesis was investigated. In vitro assays such as micro plasma-clot cultures and doublelayer soft agar cultures served as sensitive biodosimeters to determine erythropoietic and granulopoietic injuries. Day-10-5, HA/ICR, pregnant mice were irradiated with 0, 50, 100, 150,200, or 300 rads, and day-14-5 fetal livers were studied for colony-forming unit-erythroid (CFU-E), buist-forming unit-erythroid (BFU-E), granulocyte-macrophage colony-forming cell (GM-CFC), and macrophage-colony-forming cell (M-CFC) activity. Fetuses subjected to doses of 200 rads or higher on day 10-5 of gestation responded with a decrease in day-14-5 liver cellularity, reflecting injury to the developing organ and its inability to recover to the nonirradiated values. Difference in response between erythropoietin(EPO)-dependent and EPO-independent CFU-E strongly suggests existence of two populations of erythroid progenitor cells with different radiosensitivities. A dose of 200 rads markedly reduced CFUE recovery, and a dose of 100 rads was sufficient to reduce BFU-E recovery to almost 10% of 0-rad values. Nonirradiated day-14-5 fetal liver had more GM-CFC compared to any of the irradiated fetuses, and a dramatically reduced M-CFC recovery occurred with each increase in dose following 150 rads. Our results showed that (1) fetal liver granulopoiesis is more sensitive to radiation injury compared to erythropoiesis, and (2) fetal liver has a greater potential for erythropoiesis recovery.

Hemopoiesis in the splenectomized-pregnant mouse following low-dose total-body irradiation.

The effect of splenectomy (SPLX) and total-body irradiation (TBI) (50-200 rad) on virgin and pregnant mouse hemopoiesis was studied, using peripheral blood hemogram values and femoral marrow hemopoietic progenitor cell activity (i.e., CFUE, BFUE, and GM-CFC). The SPLX-maternal red cell counts and hematocrit values were lower than those of SPLX-virgin mice, reflecting the anemia of pregnancy. Bu...

In utero radionuclide exposures and risk of leukaemia

Ionising radiation is a known leukaemogen and in utero exposure to diagnostic X-rays associates with increased risk of childhood leukaemia (see abstract by R. Wakeford). This paper examines the methodology used by the International Commission on Radiological Protection (ICRP) to estimate doses to the child following in utero radionuclide exposures, considers experimental evidence for in utero h...

Effects of prenatal irradiation on fetal, neonate, and young adult murine hemopoiesis.

B6D2F1 mice received cobalt-60 radiation on day 10.5 of gestation at doses of 50 to 300 rad at a dose rate of 40 rad per min. These animals were studied at four selected age periods: (a) day 14.5 of gestation, (b) neonate, (c) juvenile, and (d) 13 week-old adult. Fetal liver cellularity, morphology, and hemopoietic progenitor cell concentration reflected injury after 200 rad. The 15 day-old mou...

Fetus, Fasting, and Festival: The Persistent Effects of In Utero Social Shocks

The Fetal Origins Hypothesis (FOH), put forward in the epidemiological literature and later flourished in the economics literature, suggests that the time in utero is a critical period for human development. However, much attention has been paid to the consequences of fetal exposures to more extreme natural shocks, while less is known about fetal exposures to milder but more commonly experience...

Early chimerism threshold predicts sustained engraftment and NK-cell tolerance in prenatal allogeneic chimeras.

The failure of engraftment in human cases of in utero hematopoietic cell transplantation (IUHCT) in which no immunodeficiency exists suggests the presence of an unrecognized fetal immune barrier. A similar barrier in murine IUHCT appears to be dependent on the chimerism level and is poorly explained by a lack of T-cell tolerance induction. Therefore, we studied the effect of the chimerism level...
